Goals and Objectives

This University of Wisconsin-Madison professional development course covers background and practical aspects of solubilization and enhanced oral bioavailability formulation strategies. The formulations considered are those used in preclinical and clinical, particularly phase 1, programs for low solubility drugs. Preformulation background in solubilization is covered for pH adjustment, cosolvents, cyclodextrins, surfactants and oil-based lipids, with an emphasis on the main excipients, their levels and how to minimize the solubility measurement workload to achieve the desired formulation. The importance of precipitation upon aqueous dilution is considered for these formulations. Nonsolubilization approaches covered are milling, particularly nanomilling, and solid dispersions. The major physical tests to assess milled and amorphous solids are examined in the context of preformulation characterization and problem solving.

Who should attend

Scientists and technical managers who work in early drug development and who are responsible for preclinical and/or phase 1 clinical formulations. Attendees should include BS, MS and PhD graduates with less than 5 years of experience in the formulation discipline. Additional scientists and managers who would benefit from the course include those working in analytical, preformulation, various biological sciences (toxicologists, pharmacokineticists, pharmacologists), and other groups interested in gaining a better understanding of principles and practices of preformulation and early formulation development.

The day begins (8:00am) with a continental breakfast and includes lunch and break service throughout the day. All classes will begin at 8:30am and end at approximately 5:00pm.

1) Solubilization approaches used for poorly soluble compounds
2) Bioenhanced formulation strategies for phase 1 capsules and tablets, including particle size reduction techniques, amorphous forms, solid dispersions and liquid-filled capsules
3) Preformulation characterization and background for solubilization, milled and amorphous solids, as an integral part of formulation development
4) Excipients used in solubilization, nanomilling and solid dispersions, their functionality and common levels

Dr. Sacchetti has 14 years of industrial experience, mostly at GlaxoSmithKline, in applying physical chemistry to the physicochemical characterization of NCEs and excipients used in oral and parenteral dosage forms as well as in developing formulations for inhaled powders. Dr. Sacchetti currently serves as Scientific Director at the Zeeh Pharmaceutical Station at the University of Wisconsin School of Pharmacy, where he oversees scientific and technical aspects of contract and grant-based programs in drug discovery and development for clients on and off the University of Wisconsin-Madison campus. He supervises Station laboratory staff, provides technical training and serves as faculty for Station-offered short courses in drug development.
